ADENOVIRUS CUSTOM VECTOR DESIGN 

Different designs can be undertaken for the construction of the shuttle (transfer) vector according to the client’s objectives. The adenovirus transfer constructions could bear one (monocistronic) or two (bicistronic) expression units and additional elements targeting a variety of purposes for the recombinant encoded product (e.g. subcellular localization or secretion of the product, protein tags for purification or detection, gene overexpression, eGFP protein). Custom adenoviruses are routinely produced at high viral yield and guaranteed fast delivery.

 

 

LARGE SCALE ADENOVIRUS AMPLIFICATION, PURIFICATION & TITRATION

Possibility to produce the recombinant adenoviruses in our complementing cell line in shake flasks, in 1L, 3L and 25L bioreactors. Different degrees of purity can be selected after cell harvesting for separation of the viral particles. The analytics for characterization of the product and quality control consist of determination of host total proteins and DNA, titer (expressed as total viral particles per mL, calculated  by anion-exchange HPLC and absorbance at 260 nm or ddPCR and infectious titer of the viral stock, measured by TCID50. The titer of final preparation is about 1 x 10^11 IVP/mL.
